Fifteen residues, four of them proline

A pentadecapeptide with the sequence Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val. Proline-rich sequences like this one are where solid-phase synthesis most often drops or duplicates a residue.

Where BPC-157 slips

Sequence errors

Mass spectrometry confirms all fifteen residues and catches the deletion and insertion products that proline-rich sequences generate during synthesis. A missing residue shifts the mass by a known amount, which is exactly the kind of thing weighing the molecule reveals and timing it does not.

Salt form and net content

This is where BPC-157 buyers are most often surprised. Material sold under different salt descriptions can carry meaningfully different net peptide content for the same stated milligram figure.

A 5 mg vial of a TFA salt at full purity holds roughly 4 mg of actual peptide. Nothing has gone wrong — that is simply what the salt weighs.

What the largest dataset found

94.2–115.1%

was the interquartile range of measured amount for BPC-157 — one of the three widest spreads in the dataset. Median amount was 103.50% of label. 3.5% of samples did not contain the stated peptide at all.

What your certificate reports

  1. Identity — confirmed from precursor-to-product-ion transitions on a triple quadrupole instrument, not inferred from retention time.
  2. Purity — measured at the quantifying transition, with the method named on the report.
  3. Net peptide content — quantified against an external calibration curve and reported as its own figure, in milligrams. Why this is a different number from purity.
  4. Peak data — the chromatographic results behind each figure, printed on the certificate rather than held back.

What if the result comes back bad?
