Forty-four residues, and a hexenoyl cap

A 44-residue analogue of growth hormone-releasing hormone, carrying a trans-3-hexenoyl group on the N-terminal tyrosine. That modification is what distinguishes it from plain GRF(1-44), and it is the part of the molecule a shortcut is most likely to omit.

What length does to a synthesis

Length is the problem

Solid-phase synthesis adds one residue at a time, and each step completes at slightly less than 100%. Over 44 cycles those shortfalls compound into deletion sequences — chains missing a residue here or there, close enough in mass to pass a careless look and wrong by any real measure.

This is why purity figures matter more for long peptides than short ones, and why we report the measured mass rather than a yes-or-no on identity.

The cheaper relatives

Sermorelin is a 29-residue fragment of the same parent hormone and costs considerably less to make. So does CJC-1295. Both are sold into the same market and both sit thousands of daltons away from tesamorelin, which makes the substitution easy to detect and apparently still worth attempting.

The missing hexenoyl group

Strip the trans-3-hexenoyl modification and you have unmodified GRF(1-44), about 96 daltons lighter. That gap is under two percent of the total mass and entirely invisible to identification by retention time.

What your certificate reports

  1. Identity — confirmed from precursor-to-product-ion transitions on a triple quadrupole instrument, not inferred from retention time.
  2. Purity — measured at the quantifying transition, with the method named on the report.
  3. Net peptide content — quantified against an external calibration curve and reported as its own figure, in milligrams. Why this is a different number from purity.
  4. Peak data — the chromatographic results behind each figure, printed on the certificate rather than held back.
